The main event is the most lopsided based on ladder position, but it is still expected to be a tough, closely-fought battle.
Brothers have a three-way share of the lead after shock losses by Kangaroos and Mossman-Port Douglas, and a win will be crucial as they jostle for top spot.
Brothers beat Ivanhoes 26-14 last week, while Tully gained plenty of momentum and confidence out of their 62-26 win against Suburbs.
Tully, meanwhile, are eighth, three points outside the top six and desperate for a win to keep within striking distance on the run home.
The reserve grade clash features two of the five teams locked in equal second on 12 competition points, with both boasting a 5-3 record.
They are both three points behind leaders Kangaroos, but the top six finals format also means the team which finishes second will also get a first-week bye and direct passage to the semi final.
In the U18s, Brothers is one point leaders Mareeba as part of a logjam in which six teams are separated by just three points.
Tully is seventh, one win outside of a finals berth and desperate for a win to ensure they stay in touch with the top six teams.
